Methylmalonic acid (MMA) in serum is measured to help diagnose a number of disorders, primarily vitamin B12 deficiency. This application note describes the extraction of methylmalonic acid from serum, prior to LC-MS/MS analysis. An effective and efficient ISOLUTE SLE+ protocol optimized for the 200 uL capacity 96-well plate format is used. The simple sample preparation procedure delivers clean extracts and analyte recoveries greater than 80% with RSDs below 10%.
